Inserm researchers at the Institut Curie have identified the cells that cause Ewing’s sarcoma. They are cells of the mesenchyme, the connective tissue that supports other tissues. The Institut Curie is the reference center in France for Ewing’s sarcoma, a bone tumor of children, adolescents, and young adults. The researchers have also succeeded “to make” the tumor cells to become virtually normal mesenchymal cells again. These results  open up new therapeutic possibilities for blocking the development of Ewing’s sarcoma in young patients.

When cells learn (again) to make fat… These are cells from a Ewing’s sarcoma. Ewing’s sarcoma (cancer of the connective tissue) is the second most frequent malignant bone tumor in France, with 50 to 100 new cases a year. It occurs in children, teenagers, and young adults (up to 30 years of age), at a frequency that peaks around puberty, between 10 and 20 years of age. This bone tumor essentially grows in the pelvis, ribs, femur, fibula, and tibia. It is highly invasive and metastases are common, especially in the lungs and skeleton.

Treatment of Ewing’s sarcoma, has progressed greatly in the last thirty years. Nowadays, the therapeutic strategy used in most cases combines chemotherapy, radiotherapy and surgery. The Institut Curie is the reference center for Ewing’s sarcoma in France, and is internationally renowned both for clinical management of patients and research into this disease.

New therapeutic leads

Cancers rarely have a simple molecular signature—a specific mutation that causes tumor growth. In the case of Ewing’s sarcoma, a molecular signature was identified and characterized in 1992 by Olivier Delattre’s Inserm team at the Institut Curie. It is an accidental change of genetic material between two chromosomes, which results in the formation of a mutant gene, which codes for an abnormal protein called EWS/FLI-1. This discovery led on to the development of a diagnostic test for Ewing’s sarcoma in 1994.

Yet until now, the nature of the cell in which this mutation occurs was unknown.

The group of Olivier Delattre, the Director of Inserm Unit 830 “Genetics and Biology of Cancer” at the Institut Curie, and the team of Pierre Charbord, the Director of Inserm Laboratory ERI5 “Microenvironment of Hematopoiesis and Stem Cells” in Tours, have now discovered that Ewing’s sarcoma are caused by cells of the mesenchyme, a connective tissue that supports other tissues. They have shown that the profile of the transcriptome* of Ewing’s sarcoma ressemble that of mesenchymal cells, particularly mesenchymal stem cells, when EWS/FLI-1 is inhibited.

By inhibiting the abnormal protein EWS/FLI-1 that causes Ewing’s sarcoma, the researchers also “forced” the tumor cells to return to their original status of mesenchymal stem cells, which can then differentiate normally into bone or fat cells. This approach opens up new therapeutic prospects, since by forcing the cells to resume their original function it may be possible in the future to make them less aggressive and prevent their proliferation. As long as the tumor cells are still able to fulfill their function, they generally proliferate slowly, and the prognosis is good; once they lose this capacity, however, the tumor cells become highly aggressive.

This discovery could allow Delattre, Charbord and colleagues to produce an animal model of Ewing’s sarcoma, an essential stage in the development of new treatments.  These results were published in the May 7 issue of Cancer Cell.

*The transcriptome comprises all of a cell’s messenger RNA, which carries information from part of the genome for the synthesis of proteins.

Steven Mackin w/ Wonder Woman PosterYesterday’s balloon launch was indeed a celebration, and also a day where we could come together in Steven’s memory and pay tribute to him.  I went home and crashed out on the bed, and went to sleep until late today – and I had something on my mind.

Of course, if you speak with Steven’s family they will tell you that Steven’s passing has brought them close to people from all over, in many special ways.  There is fate working it’s magic here, and that is all that they (and I) can say regarding this project.  How do I mean fate?  Well, lets see – here is one really interesting example: October 28th was recently named “Wonder Woman Day,” in Portland, Oregon.  Steven was a big “Wonder Woman” fan, see link below for the actual proclomation!

Of course, October 28th was the day that Steven passed away – and I think that in some way, (call me kooky) Steven had this day dubbed “Wonder Woman Day,” to give us all a smile and a wink-wink-nudge-nudge.  I can see him now laughing about us all figuring this out.  Wonder Woman should be so proud, huh? ;)

 There are many others, some are far too personal to be put on a blog like this – but you get my drift.  In many many ways, Stomp Out Cancer was given a little help from above, from a big personality – and we can’t thank you enough for your support!

The Kentucky Standard 10/28/07

Kentucky Standard Small

The most beautiful article was written about Stomp Out Cancer, the balloon launch, and the CD release today by “The Kentucky Standard,” a wonderful newspaper in Bardstown, Kentucky.  The article captures the project, Steven’s life, and advertised the balloon launch (which we will have photos up of soon) from today.  The CD’s sold, people released dozens of colorful balloons, and I got to send up my set of balloons along with M.D. Cunningham (artist from the CD, and friend of Steven’s) – which really was special to me.  Steven’s family was there and I got to meet so many new wonderful people!  The day, while sad in many respects, was definately a celebration of Steven’s life – and the memories of him.

Stomp Out Cancer: Celebration Balloon Launch

Stomp Out Cancer CD Release PosterThe Stomp Out Cancer benefit compilation CD is due to be released on Sunday, October 28, 2007. This is a very special date, as it is the one year anniversary of the passing of Steven Michael Mackin, the young man who this project is dedicated to. In conjunction with album release and in honor of their son, Steven’s family is planning a very special celebration. On October 28th at the St. Joseph’s Cemetery in Bardstown, KY, gatherers will launch balloons at Steven’s grave site carrying messages to him in celebration of his memory. “We are hoping that people will be able to come with their balloons and fill the sky with color, just as Steven filled the world with his warmth for so many people all over the World.”

CD’s will be in limited supply at the balloon launch, but you can obtain an order form to either order online, or mail in payment to have yours shipped.

The balloon launch will start at 2:00 pm est., and everyone is welcome to attend. “We just ask that you please BYOB (bring your own balloons) and attaching a message is optional. “People may even want to remember another loved one who passed from cancer, and that is ok too. This is a celebration of our love for our son, and a day to simply remember how great he was.” Sheila explained.

Stomp Out Cancer: Indie Artists Fight Ewing’s Sarcoma is a compilation CD filled with beautiful tracks of hope, love, and memorial will also be released to raise money for less toxic treatment methods, and early detection methods.

“This CD has been in the planning stages for months, and we are so proud to finally see it being released. The music is from various artists who have lost loved ones to cancer, or who related to this project in some way. All proceeds from this CD will go to help fund research that is not funded by the FDA, because they say that it is too rare.” Sheila stated. Ewing’s Sarcoma is found mostly in children and teenagers, and currently the FDA does not have any funding for research into this disease. One person a day is diagnosed, but according to the FDA, this is simply not enough.

I am writing to first express my admiration and support for the work you are doing.  For over 15 years I have been caring for patients with Ewing’s sarcoma and conducting research on this disease.  While progress has been made, there is much more work to be done and too many wonderful lives are lost to this disease every year.  I am excited to get my CD and share it with our community at NIH and I look forward to discussing potential options for research funding.  The focus of our research is to develop immune therapies for Ewing’s sarcoma, which we believe will be less toxic and  better able to control this disease in the long run.
